Dornberger et al. (2026). Diabetes, Obesity and Metabolism.
Summary
A multicentre retrospective case series of 20 adolescents with obesity related to craniopharyngioma, a brain tumour, treated off-label with semaglutide. Before treatment their BMI was rising by about 3.5 kg/m2 a year; after one year of treatment it had fallen by 2.5 kg/m2 (p<0.001), and 40% had a BMI reduction of 10% or more. Gastrointestinal side effects occurred in 30%, all mild. There were no discontinuations, pituitary decompensation or tumour recurrence. It is a small uncontrolled case series with no comparison group.
