Lipidated macrocyclic peptide S-880008 inhibits SARS-CoV-2 fusion by suppressing protomer dissociation at conserved spike sites

Sanaki T, Kawaguchi Y, Anraku Y, et al. (2026). Nature Communications.

Summary

A laboratory and mouse study of S-880008, a lipid-tagged macrocyclic peptide. It binds conserved sites on the SARS-CoV-2 spike, holding the spike protomers together and blocking viral fusion, with antiviral activity in vitro against variants including Omicron. Given intranasally to adult mice infected with SARS-CoV-2, it lowered viral titers in the lung and improved survival. This is a cell and mouse study.
