GLP-1R agonist semaglutide protects human forebrain organoids from cholesterol-induced neurotoxicity
Yu et al. (2026). Alzheimer's & Dementia.
Summary
A laboratory study using human stem-cell-derived forebrain organoids exposed to excess cholesterol, a model of lipid-driven neural injury. Semaglutide protected neural cells from the injury and was linked to activation of cAMP-PKA and PI3K/Akt-mTOR signalling, less lipid droplet build-up and less oxidative stress. Single-cell analysis suggested partial preservation of disrupted cell-to-cell signalling, including the midkine network, and calcium activity patterns were stabilised. This is a cell-model study with no animals or people. Funded by Chinese national and provincial research grants; no industry funding disclosed.
