Retatrutide, a Triple Hormone Receptor Agonist, for Treatment of Obesity

Jastreboff et al.; TRIUMPH-1 Investigators (2026). New England Journal of Medicine.

Summary

The phase 3 TRIUMPH-1 trial randomised 2,339 adults with obesity to retatrutide, a GIP, GLP-1 and glucagon receptor agonist, at one of three dose levels or placebo for 80 weeks. Body weight fell by 17.6%, 23.7% and 25.0% in the retatrutide groups against 3.9% with placebo. Among 574 participants with knee osteoarthritis, pain scores improved more than on placebo, and among 243 with obstructive sleep apnoea, the apnoea-hypopnoea index fell. Gastrointestinal events were the most common adverse events. The trial was funded by Eli Lilly. Retatrutide is investigational and not an approved medicine.
